Every ENFINIA sequence—whether Linear DNA, IVT-Ready DNA, or Plasmid DNA—undergoes a stringent quality control protocol before shipping. We evaluate every construct across three critical dimensions:

  • Yield: Quantified using high-sensitivity fluorescence and spectrophotometry assays to ensure the sequence meets or exceeds the specified yield.
  • Identity: Verified via long-read NGS sequencing to ensure a 100% match to your reference sequence, with zero SNVs.
  • Purity: We maintain strict internal thresholds to minimize side products.

Long-Read Sequencing for Full-Length Resolution

Elegen uses industry-leading long-read NGS platforms to analyze entire native DNA molecules. Whereas short-read technologies require DNA fragmentation, ONT can sequence multi-kilobase molecules in a single, continuous read.

Validation Beyond the Building Blocks

Long-read sequencing provides true end-to-end verification. Unlike traditional workflows that only assess precursor fragments, we validate every customer sequence in its final, full-length form.* This eliminates the risks of “discontinuous” QC and identifies assembly artifacts that other platforms often overlook.

Furthermore, the NGS verification platform resolves complexities—such as hairpins, high-GC content, and repetitive elements—that exceed the physical limits of short-read sequencing. By spanning the entire construct, our NGS-verification ensures every element is accurately placed within its proper genomic context, resolving the structural ambiguities common in fragmented data.

While other providers limit NGS to clonal plasmids, Elegen provides comprehensive characterization for every sequence we ship, regardless of length, complexity, or format.

*For IVT Ready DNA, poly(A) tails undergo strict quality assurance, and the presence of the poly(A) tail on customer sequences is confirmed, but the specific tail length on each sequence is not quantified.
