Using ENFINIA™ Linear DNA in Gibson Assembly Reactions

Since it was originally introduced in 2009, the Gibson assembly method has been widely adopted by the research community. Among in vitro DNA assembly methods, it is one of the most favored for its simple one-step protocol and ability to generate scarless, recombination-based constructs.

In this application note, we demonstrate that ENFINIA Linear DNA can be used directly in Gibson Assembly reactions without prior adapter removal. This capability not only preserves the integrity of the synthesized construct but also significantly reduces processing time, lowers costs, and simplifies the cloning workflow.
